No. Grant and Lee knew each other from West Point and the Mexican War, however Lee's brilliance was recognized then. Lee was offered top General of the Union right before the Civil War broke out, however he politely declined it. After going through a half dozen Generals, Lincoln gave Grant a chance and he proved to be a capable General.
